摘要:sonicationisasimplemethodforreducingthesizeofliposomes.wereportthesizedistributionsofliposomesasafunctionofsonicationtimeusingthreedifferenttechniques.liposomes,mildlysonicatedforjust30sec,hadbimodaldistributionswhensurface-weightedwithmodesatabout140and750nm.withextendedsonication,thesizedistributionremainsbimodalbuttheaveragediameterofeachpopulationdecreasesandthesmallerpopulationbecomesmorenumerous.independentmeasurementsofliposomesizeusingdynamiclightscattering(dls),transmissionelectronmicroscopy(tem),andthenystatin/ergosterolfusionassayallgaveconsistentresults.thebimodaldistribution(evenwhennumber-weighted)differsfromtheweibulldistributioncommonlyobservedforliposomessonicatedathighpowersoverlongperiodsoftimeandsuggeststhatadifferentmechanismmaybeinvolvedinmildsonication.theobservationsareconsistentwiththefollowingmechanismfordecreasingliposomesize.duringultrasonicirradiation,cavitation,causedbyoscillatingmicrobubbles,producesshearfields.largeliposomesthatenterthesefieldsformlongtube-likeappendagesthatcanpinch-offintosmallerliposomes.thisproposedmechanismisconsistentwithcolloidaltheoryandtheobservedbehaviorofliposomesinshearfields.
